Hope all is well. My sister's study visa has been approved. She is currently in India and haven't traveled to Canada yet (so no Study Permit on hand).
She has 2 kids, 4 years and 9 years old. Can she apply for her kid's study permit now or she has to enter Canada and have her Study Permit before applying?
If it's possible to apply, what explanation should we use for "Letter of Acceptance" since they don't have school acceptance letters?

LOA is not required for minor dependents. Just upload a letter explaining that school is to be determined depending on local school board. Can apply for minor’s SP now together with PA parent. Or you can apply for TRV now. Others were able to study in primary and secondary schools with just a visitor permit and minor dependent of international students.

Your 4 year old needs a TRV while the 9 year old needs a study permit. Previously, the 9 year old could also apply for TRV but IRCC's policy on this have changed. All school age kids needs to apply for a SP now, otherwise their application gets returned/cancelled and they will be asked to apply for a SP.
You can apply now but not certain she will get it in time she is planning to go with them for fall. There are no guarantees that they will approved either since she didn't include them in the application from the onset.
Your 4 year old will not need a study permit even to attend private school. You can however still apply for SP for them if you'd prefer, whether they are attending private/public school. It's just an unnecessary additional expense in my opinion.
As other members clarified, below is the link and table that will guide you. In your sister's situation, Parent SP was approved before arrival to Canada, then Minor don't need SP to go school, and they will be issued Case Type-10 Visitor Record at Point of Entry.
