I got three-year PGWP and expired in Aug 2019. I applied one-year post-graduated program as a second degree, and hopefully will get ITA upon graduation in Apr 2020. Would I be able to qualify regular work permit application after graduation and also assuming ITA is issued luckily? With my understanding, I can't apply BOWP and PGWP again. Any advice and successful story with regular work permit application?

You will only qualify for a regular work permit after graduation if you find an employer willing to give you a job offer and if your employer obtains an approved LMIA.
You are correct - you cannot apply for a PGWP again.
You will only qualify for a BOWP if you are holding a valid work permit. To get a work permit, you need a job offer and approved LMIA.
An ITA will not help you get a work permit. It's completely irrelevant.
As said above, you need to find an employer who's willing to go through the process to get an approved LMIA.
