If I want to change the conditions on my current TWP, Can I go in person to Buffalo or any other US Visa office in person? Basically I want to change location in my current WP, so I think I will need new WP which can be issued in person at Buffalo, but I want to know whether I can go to any US office or not?

Send your application to CPC-V, once you found a new employer. If you currently have a valid work permit you can move to any province/location in Canada, look for an employer, apply for a new work permit or go to the nearest POE and obtain a new work permit, before you start working for the new employer.
So you need a job offer/contract from a new employer; send application to CPC-V. Or if your TRV is still valid, you can try the nearest POE and apply from there.

Are you sure this is possible, from what's written on the CIC website, only trv-exempt citizens can apply for work permits at a port of entry. Otherwise, why would anybody wanna wait 30 days to renew his work permit, when he can just go at the borders an do it on the same day?
