No, has to applied for at an overseas office.
Can she stay at Canada, but sent the application to the Buffalo office for example at the same time?

Yes, you can apply from within Canada to extend your parent's stay in Canada through the Super Visa. You send the application to Vegreville, AB if it is just for the extension of visitor status. However, if your mom plans to leave Canada and then come back, then she would need to have the Super Visa stamped into her passport. In that case, you have to send the application to Buffalo, NY.

I was told by the CIC agent that I could also apply online. However, this would require scanning all the documents, including insurance certificate, insurance policy, etc. I still preferred sending in a paper application even though online application is much faster. Remember that your online application is still submitted to Vegreville, AB. That's what you chose in your online application, right? Best of luck!
An application for a new single- or multi-entry visa must be done from outside Canada at a visa office.
However, parents and grandparents who are in Canada on an authorized stay of up to six months may apply for an extension of their status of up to two years by submitting an application to the Case Processing Centre in Vegreville, Alberta, as long as they meet all the criteria for the Parent and Grandparent Super Visa.
