Staying for 3 days in Vancouver before entering final destination Regina.

I recently received COPR letter and VISA for Regina, Saskatchewan under PNP program of Saskatchewan. On my COPR, it is written that my province of destination is Saskatchewan and City of detination is Regina.
Now I want to book my flights for Regina. But since there are no direct flights, I would have to go via Vancouver, Toronto or Calgary.
Since I have my friends staying in Vancouver, can I take a stop of two three days at Vancouver before taking my connecting flight to Regina. I will book my tickets from Vancouver to Regina in advance to show it to the immigration officer at Vancouver airport during my immigration process.
