Provinces are naturally selfish of the immigrants that they bring in because it costs them money to bring in quality talent and they would appreciate if the talent isn't just using them to get entry into Canada. They would also be keen on the immigrant talent settling down in the province to add to the provincial tax kitty. Which explains the insistence on getting the declaration of intent signed upfront.
However, the constitution always has the last word.
Notwithstanding anything else to the contrary, The Canadian Charter of Rights and Freedoms allows the following mobility rights to citizen and PR.
6.(1) Every citizen of Canada has the right to enter, remain in and leave Canada.
(2) Every citizen of Canada and every person who has the status of a permanent resident of Canada has the right
(a) to move to and take up residence in any province; and
(b) to pursue the gaining of a livelihood in any province.
(3) The rights specified in subsection (2) are subject to
(a) any laws or practices of general application in force in a province other than those that discriminate among persons primarily on the basis of
province of present or previous residence; and
(b) any laws providing for reasonable residency requirements as a qualification for the receipt of publicly provided social services.

Is there any application quota for SINP-Express Entry sub category? And is there any source from where we can know that how many applications have been accepted under this stream?

Yes, 1000 applicants only will be nominated under SINP Express Entry. The source where you will get the status - http://www.economy.gov.sk.ca/immigration/sinp-application-intake-thresholds. This is not realtime status and SINP does not update it every now and then, so it does not reflect the actual number of applications already filled towards the quota. SINP started on 08 June 2015 and are currently still accepting applications.
