I worked part time as a Technical support Analyst for 2 and half years (2282) (approximately 16 hours per week) while doing my B.tech. But When I was applying for my study permit,I didn't mention this.
So, My question is Can I use this experience for my Express entry profile now? I have the experience letter and pay stubs.
Any work experience accumulated while studying full time is NOT valid.

This is only true if in Canada. Outside of Canada, work experience while studying is accepted.

But what if I didn't mention it while I filled my application for study permit (3 years ago)?
