Yes - your US overstay will impact your ability to get a visa for Canada. Any sort of temporary visa (visitor visa, work permit, study permit) will be refused. If you want to come to Canada, you will need to focus on applying to immigrate and become a PR.
what do you mean by Canada spouse visa? Is your spouse a PR or Citizen? If so they can sponsor you and you will not be rejected for your overstay. If your spouse is in Canada on a temporary basis and you want to visit them then your overstay in the USA could negatively impact your file.
Canada and the US share immigration records. Canada will know.
You will also be asked about your immigration history in the application to Canada. If you lie about your overstay, Canada will find out about it anyway through the record sharing with the US and then you will be refused and banned from Canada for 5 years.
I'm not sure what you are asking. The fact you enter the US on a transit visa doesn't change anything about your situation. You are in the US illegally and have been in the US illegally for many years now. Again, any temporary visa to Canada is not possible for you. US immigration records will show that you entered the US and never left. These records are shared with Canada and Canada will know about your overstay.
