Having graduated from a public university with a bachelor's degree, I have now obtained one year's full-time work experience.
Soon I will file the application for CEC immigration.
Since I already have the 1 year work experience and have satisfied all other requirements, I want to quit my job and go back to school to get more education.
So I am wondering if it would be an issue if I quit my job after I file the application. Does Immigration Canada require applicants to secure/have a job when they assess the application? Does "not having a job/not working" affect the application process or cause any delay in any way?

Nope. If you are 100% certain you have met the work requirement, you can quit and go back to school. The only thing you need to be concerned about is maintaining your legal residency, having all the right permits/visas, etc. And good luck!
