I have an urgent query.. While landing first time, do we really need to fill and Carry Form "IMM 5444E" with us or Immigration officer can fill at the port of entry.

As per my experience you don't have to. The immigration office who makes you sign the COPR applies it on your behalf at the time of landing, although I read in the forum that you need to carry a filled form. You just need to provide her your Canadian address (which you will have it anyway ready on your Good accompanying or Goods to follow list).
I landed in Toronto with my family and the immigration officer went ahead and applied for our PR cards (as it is part of the landing procedure itself).
