I had my mother's Medical done for Super Visa 2 days ago. Should i wait for IRCC to request medical for PGP application or just submit it via email with 5768

Submit if available recent pcc and medical to short the process time in case they accept will be good, no harm to share.

I got request for additional docs on 22 Feb and submitted the docs by email on Mar 01.

what does "all dependants aged 18 and over, whether accompanying or not.:" means? my brother who is already married and not part of the application should also complete the forms?

No. Because he is not a "dependent". The instruction state all dependents aged 18 and over. (i.e. in general children that's 18 - 21 that's applying under their parents as part of the PGP application)
