Yesterday I changed the answer to 'Does DY have a job offer in Canada?' to NO and my profile has been updated/transmitted
Still NO new message/pdf file indicating Job Validation Code,
Therefore cannot apply for PNP.
What should I do? Should I just withdraw and start a new one?

Better to withdraw the current one profile and create a new one by saying No to job offer question.
when you update any information in EE you get a message on your email that your EE has been updated after 24 hours and you don't get a new PDF document/message in your inbox for that , it just updates your profile online.so i guess for that create a new one
