Does he have a common law partner or children? Are either of his parents still alive? Does he have any other family in Canada (e.g. aunt, uncle)? If the answer to any of these questions is yes - he unfortunately cannot sponsor his sister.
Oh - and to answer your other question - if he qualifies to sponsor his sister now and applies - he cannot get married until his sister's PR application has been finalized and she has landed in Canada and become a PR. Sibling applications can often take 2-3 years to process. So he will need to be prepared to wait a few years before getting married. Once he is married, he will be able to sponsor his spouse for PR.

In that case he cannot sponsor her. His sister will have to qualify to immigrate on her own through an economic immigration program like Express Entry.
If you live in Manitoba, she will get more points to qualify for the Manitoba PNP program. However she will still need to qualify on her own as an economic immigrant and meet all of the education, work experience and language requirements. This isn't a family sponsorship stream - it's still an economic stream.
