Do you have any family in Canada?

I believe it means that you have to be the following:

- single (not married, not in a conjugal relationship, do not have a common law partner)
AND
- do not have Any relatives in Canada (whether they are Canadian citizens or Permanent Residents).

Yes they are alive, and live in Pakistan.

You may sponsor your parents as a member of the family class, so for that reason you don't qualify to sponsor your brother.
