My wife sponsored me into Canada. I live with her and our 2 dependent children (4 and 7). I hold a valid work visa and have completed all my medical exams. My wife is currently dealing with health and mental issues. I have a job and I am supporting them. If she was to go on Government assistance would it cause any issues to our PR application?

Yes - it would impact the application. She cannot be on social assistance to sponsor you. If she takes social assistance, you should expect the application to be refused.

It will likely still be a problem. She is responsible for you financially for the first three years after you become a PR. If you take social assistance during this time, she will have to repay this money to the government. Since you are married, I don't believe it will be possible for her to go on social assistance herself, you would be included in the application.
Are you working? If so she may not qualify for welfare. We may not qualify for disability either. Depends on whether you have savings and your family income. You may need to get a second job.
