My Wife is applying for Canada student visa to which I am certain she will get it. I am sponsoring her (school fees, her stay and other expenses). My question is, do i have any chances of getting an open work permit?
I am running a transportation company which is currently doing very good.i have other assets as well back home so strong ties back home is not an issue as well.

Yes, your chances are very high. Canada tries not to do anything to separate families. So, they will give you whatever it would require to keep you together. Please do not apply together with her. Once her SP is approved, you can apply for a visit visa and include a copy of her permit and visa. If you apply together, they may suspect you want to emigrate cheap. Just get a visit visa, and upon arrival in Canada, change your status. You have a very high chance of approval of an open work permit for you, provided your spouse is actually registered in a publicly-funded institution in Canada. Applications made while you are in Canada have a far higher chance of success than the one made from your country.
