Can anyone have an idea about the occupation 4121 (university Professor) is it required to hold PhD or not?
because my job title is University Lecturer but I dont hold PhD just MSc (Master Degree).

It says at http://www5.hrsdc.gc.ca/NOC-CNP/app/quicksearch.aspx?lc=e&searchkey=4121 that a doctoral degree is required.
 
No, CIC ignores the professional requirements when evaluating an application.

What you have to ask yourself is:

- Does my job fit the "lead description" (the first paragraph in the NOC)?

- Do I carry out most or all of the duties specified in the NOC?

I checked the website of CIC to know if I am qualified or not under Skilled worker program and I answered as follow:

* You have not completed post-secondary studies or worked in Canada.
* Your experience is in the “none of the above” category.
* You have skilled work experience.

then if I answer yes for this question:
Has an employer in Canada given you a full-time job offer* in writing?
then it is apparently that I am qualify ( the printed message was)
"Based on your answers, you may be able to immigrate under the Federal Skilled Worker Program.. "

The question is there any condition on the job offer or just a letter said that we offer ..... a job in ..... company.

For a skilled worker job offer, the job has to be classified as skill level 0, A or B under the NOC code and the employer needs to apply to Service Canada for an AEO = arranged employment opinion.
