I have a unique case. I am sponsoring my daughter. Her mother and I were never married and my daughter had been living with her mother so far. Now we all have agreed that I can bring my daughter to Canada. Her mother has her own family now (basically her own husband and children). To prove our relationships, embassy had asked for our DNA samples. I gave my DNA samples from Canada few months ago and now they have scheduled an appointment for DNA sample collection for my daughter and her mother in New Delhi. But..
The main problem is that my daughter's mother is not willing to go and give DNA samples. She is kind of stubborn type of person and as far as I know her she wont go to the appointment to give her DNA sample. So, is it going to be enough that CIC verifies our father-daughter relationship? Or, not getting DNA samples from the mother can be a reason for rejection of the application? (Its not our fault that the mother does not want to give DNA samples .) What can be my options?
Since only the "real" mother can approve the child's move to a foreign country, I'm betting your application is in for a LONG haul (even stalled permanently) until the requested information (verifying she the real mother) is submitted. I'm eager to see however what a legal expert here will have to say on this matter.
I had the impression that just from the sample of one parent, they can determine if she is your child or not (I dont think they need both), however that was just the impression I had.
Maybe the mother could sign a letter stating that she does not want to give a DNA sample and state why, otherwise be open with CIC and explain to them the situation and show that you've done everything in your power to get that, but there is so much you can do
Thank you for your response truesmile. The real mother has already approved and signed paperwork for me take her with me. We have already signed a custody agreement with a lawyer. But now she is being a pain. Its just the DNA test she does not want to do.

I'm not sure I understand what you mean,
But what I meant is, as a last resource: make sure to show (in a letter) to CIC that you have done everything in your power to get the DNA sample and that she has been uncooperative. Ask what other possibilities you have to prove that you are related, dont just say: "hey, I cant get you guys the DNA sample" at the end of the day, they are humans, and they deal with so many cases that most likely you are not the first person to have this problem, so chill ;D
