Yes, it is mandatory that you work for your employer in Canada for a year first to claim the 50 points. This assumes you are on a closed work permit with your employer named and not something like a PGWP.

isn't it come under below category.Because i cannot see the 1 year requirement in there.

You’re currently working in Canada in a job that is exempt from a LMIA, but it is not under an international, federal-provincial agreement or because of significant benefit to Canadian interests.
An employer other than the one you are currently working for:
