Here is the thing, we have applied for spousal sponsorship+OWP in June 2015. Already received the OWP. So my question is that would it be possible for her, if me and her leave the country and come back if she applies for multiple entry visa?
There is no guarantee she will receive a multiple-entry visa. If she needs a visa to enter Canada, she should apply for it before she leaves Canada. If she cannot, I would not risk leaving the country. If she is refused a visa, or refused entry, her inland application is over.

If she has an OWP, that gives status in Canada doesn't it? Wouldn't the visa be redundant? Still, leaving during an inland application is a bad idea all around.

OWP (all work permits actually) say in big.letters on them that they don't guarantee entry into into Canada or status. They just authorize you to work.
