Bio metrics updated today for me and BIL requested for my wife back home in India.

IMM 5406 Additional Document form also requested. Should we fill up 2 forms? Me as an applicant and she as an applicant? How do I upload it for both of us if its only asked for me?

Are you the PR/Citizen whose sponsoring your wife? If so, only your wife is the applicant. And only she needs to fill out this form.

I applied under CEC Express entry on September 19, 2020. My wife is back home in India and I am the primary applicant and she is my dependent spouse applicant.

Then you posted your question in the wrong section.
Because you are not sponsoring her as a family member. You are including her as part of your application.
My guess is that both of your need to supply that form (2 forms). But I do not have experince with CEC Express Entry.
