Can an American green card holder apply for a Canadian work permit?

Not directly.

You would first need to find an employer willing to offer you a full time job in Canada. If the job falls under NAFTA, you could then apply for a work permit tied to that specific employer. If the job does not fall under NAFTA, the employer would need to obtain an approved LMIA before you can obtain the work permit.
