I need some suggestions here. My current crs score is 323. I am working with an employer(from Ottawa - Ontario) for Job offer and LMIA under NOC Code 2171.
I will be adding this LMIA and job offer in my express entry profile which will be adding with 50 points. In total my CRS will be 373.
How the ITA works ? Will they process ITA for profiles with Job Offer quickly ?
E.g If there an EE Profile with 47o points(without Job offer) and another EE Profile with 375 points(valid LMIA Job offer). which will be considered first ?
Are there chances of getting ITA with 375 Points along including valid Job offer ?
So your score is 375 with a LMIA. That will not get an invitation. Do you have an approved LMIA from the employer. If so, apply for a closed work permit and see if gets approved.

You need to go through the PNP route for getting an ITA - just the job offer will not be sufficient. Also, having a job offer does not speed up or reduce processing time for EE applications.
My employer will be initiating LMIA under global talent stream which seems to be processed quickly. Apart from the Job Offer with LMIA, is there any other specific requirements for closed work permit ?

You still need a work permit. First your employer needs to determine if their job is LMIA or LMIA exempt. And if they are within the GTS. Then you apply for a work permit. Even under GTS, processing right now takes months. Their HR department or lawyers will be doing this prior to you applying for a work permit. https://www.canada.ca/en/employment-social-development/services/foreign-workers/global-talent.html
Since you are LMIA exempt, you need to work for your employer in Canada for a full 12 months before you will qualify for the 50 job offer points. So you have another four months to wait.
