Need your help. Recently I have received the passport request and accordingly send the passport and photos to the VISA office at Singapore through visa application centre. However, in my cic account, there is a request to replace digital photo and passport for me but nothing is required in my spouse's side.
Now my query is, do I have to upload again the photos and passports of me and my spouse in my cic account or else I will only wait for getting back the passport.

Thats weird. Did you get two separate passport and photo requests?
One from your local VO and another one from my cic to replace the passport and photo in your my cic account as well?
If you just received one request, then that is simply your passport request for your COPR papers and you wont have to update anything in my cic for that.
