I'm currently working full time as Mobile Security Supervisor in Toronto. I started working on Sep 17, 2017 and I went on vacation for 3 weeks in the month of April 2018.
I'm having a fixed schedule of 84 hours bi-weekly, and as per criteria I have completed 1 year and required 1560 hours as well.
Should I create my express entry profile now as it's September 2018 or wait for another month ?

As far as the anniversary is concerned you can create your profile tomorrow or after to qualify FYI for the next rounds of invitation(around 19 Sep)...

Based on the current trend, I may get an ITA so, should I create my profile on Sep 17?

During these 3 weeks you were on the payrolls on the company right ? These 3 weeks were part of your standard company benefits e.g. vacation days etc ?

In my case I joined tre organization on 27 Feb’17.... so technically my 1 year gets completed on 27 Feb’18. My EE profile was created on 16 Feb’18. I was awarded the points for CEC and the also additional points for the skills transferability.
Additional points got added later after a week for spouses IELTS as well.
Got ITA on 14 March’18 submitted application on 6 April’18.
Current status is Eligibility is Passed...
so my opinion you don’t need to wait until 17th to make a profile...

Sorry, I don't know the answer to that. Usually, unless it's in the checklist or specifically requested, don't submit things. IRCC will already have a record of it anyway.
